Users losing the ability to access mapped drives or browse to file shares

I have Server 2008 with Service Pack 2....32bit.  I have about 75 - 100 users that connect to it for file access and print.  For some reason during random parts of the day users will lose the ability to access their drive mappings.  They are also unable to browse to any of the volumes.  This starts out with just a few users then affects everyone within minutes.  Although you can ping and access the server through RDP just fine.  After a reboot of the server everything is fine.  This is happening 2 to 3 times per week.  There is nothing in the event logs that explain this.  ANY suggestions will be much appreciated.
